  Whenever I am choosing a new single product and inserting a new item, product variation like Color, Size & Brand is not getting displayed in preview and on the website.
  
  Also, I am unable to choose a desirable layout for Single products.

  The issue because you turn on catalog mode. Please go to Claue > Theme Option > WooCommerce > General Setting > Enable Catalog Mode > Off
